ABSTRACT

Objective@#To investigate the expression of syndecan-1 and syndecan-2 and their clinicopathological significance in patients with gallbladder squamous cell (SC)/adenosquamous carcinoma (ASC) and adenocarcinoma (AC).@*Methods@#A total of 126 patients with SC/ASC (n=46) and AC (n=80) were included in this study. The expression levels of syndecan-1 and syndecan-2 were detected by Envison™ immunohistochemistry assay. The clinical and prognostic significance of syndecan-1 and syndecan-2 were analyzed.@*Results@#In the 46 SC/ASC samples, syndecan-1 and syndecan-2 were positively expressed in 29 (63.0%) and 28 (60.9%) tumor tissues, respectively. (Positive expression was defined based on the staining in the component of squamous cell carcinoma. That is to say, the tissue which adenocarcinoma part was positively stained, but squamous cell carcinoma part was negatively stained is also regarded as negative.) In the 80 AC samples, 47 (58.8%) cases showed syndecan-1 positive expression, and 51 (63.8%) showed syndecan-2 positive expression. There was no significant difference in the positive rates of syndecan-1 and syndecan-2 between SC/ASC and AC groups (P>0.05 for all). The levels of syndecan-1 and syndecan-2 were associated with tumor size, TNM staging, lymph node metastasis, invasion of adjacent tissue, and surgical procedures in SC/ASC patients (P<0.05 for all). However, their expression was associated with tumor differentiation, tumor size, TNM staging, lymph node metastasis, invasion of adjacent tissue, and surgical procedures in AC patients (P<0.05 for all). The Kaplan-Meier survival analysis of SC/ASC and AC patients revealed that the average survival time for patients with positive syndecan-1 and syndecan-2 expression was significantly shorter than that of those with negative expression (P<0.01 for all). Cox multivariate analysis indicated that syndecan-1 and syndecan-2 expression were independent unfavorable prognostic factors for SC/ASC and AC patients (P<0.05 for all).@*Conclusion@#The syndecan-1 and syndecan-2 expression are associated with the tumor progression and poor prognosis in patients with gallbladder SC/ASC and AC.

ABSTRACT

Objective@#To investigation HER2 status in gastric adenocarcinoma of Chinese and contributing factors to the HER2 expression. @*Methods@#HER2 status of 40 842 gastric adenocarcinomas and clinical data were retrospectively collected from 23 hospitals dated from 2013 to 2016. The association between HER2 positivity and clinicopathologic features was analyzed. @*Results@#Of the 40 842 patients the median age was 62 years, the male female ratio was 2.6∶1.0. The rate of HER2 positivity was 8.8% (3 577/40 842). HER2 expression was related to the tissue type, tumor location, Lauren classification and tumor differentiation (P values: 0.009, 0.001, <0.01 and <0.01, respectively). Different HER2 expression status was observed between primary and recurrent tumors in 7.6% (48/635) cases. The rates of HER2 positivity ranged from 2% to 10% among different institutions. The rates of HER2 FISH amplification were dramatically different among the 23 hospitals (0-100%) with an average rate of 10% (810/8 156) in patients with HER2 IHC 2+ . @*Conclusions@#HER2 expression is associated with clinicopathologic characteristics. HER2 re-assessment of tumor tissue and use of in situ hybridization techniques increase HER2 positivity. The current retrospective study should reflect the HER2 status in gastric adenocarcinoma of Chinese patients.

ABSTRACT

Objective To study the feasibility and efficacy of prevention and treatment of rethrombosis with an implantable pump of the portal veinafter liver transplantation.Methods The clinical data of adult patients who underwent orthotopic liver transplantation from Feb.1999 to Dec.2007 were analyzed retrospectively.Results The 404 orthotopic liver transplantation (LT) patients were divided into three groups:PVT patients with an implantable pump (PVT + implantable pump,n =28),PVT patients without an implantable pump (PVT,n =20),and patients without pre-existing PVT (non-PVT,n =356).The following parameters in the 3 groups of patients were compared:(1) pre-operative parameters including baseline data of the donors,recipients,and duration of graft ischemia;(2) intra-operative and postoperative parameters including operative time,red blood cell and plasma transfusion,platelet concentrate transfusion,amount of bleeding,primary graft malfunction,and durations of hospitalization and intensive care unit (ICU) stay;and (3) follow-up data on patency of portal vein,rethrombosis rate,stenosis and reoperation (relaparotomy or retransplantation),in-hospital mortality,and actuarial one-year survival rate.Results among the 3 groups of recipients,there were no significant differences detected in the pre-operative and intra-operative parameters.However,when compared to PVT patients without an implantable pump,PVT patients with an implantable pump showed a remarkable reduction (P < 0.05) in post-operative hospital stay [(21.7 ±8.9) vs.(26.5 ± 6.1)],rethrombosis rate (7.14% vs.30%),reoperation rate (0 vs.25%),and in-hospital mortality (3.57% vs.25%).Conclusions Patients with pre-existing PVT had a higher risk of rethrombosis following liver transplantation which was significantly prevented and/or treated with an implantable pump.Implantable pump placement should be considered for PVT patients undergoing liver transplantation.

ABSTRACT

Objective To investigate the expression of DcR3 mRNA in cells of acute leukemia(AL) and its clinical significance.Methods The DcR3 mRNA of bone marrow mononuclear cells(BMNC) in the patients with AL was detected by RTPCR.There were 28 AML cases,and 18 ALL cases.27 cases of non-malignant hematopathy were used as control group.The expression level of DcR3 mRNA in BMNC was detected with RT-PCR.Results The positive rate of DcR3 mRNA was 67.4% and gene expression level was 0.56?0.09 in AL group,while they were 40.7% and 0.39?0.19 in control group,there were significant differences(P0.05).The positive expression level of DcR3 mRNA increased when the white blood cell count was ≥30?109?L-1.Conclusion DcR3 gene might play a role by suppressing apoptosis in leukemogenesis.

ABSTRACT

Objective To investigate the effect of IGF-I on the apoptosis and the expression of survivin through the signaling pathway of PI-3K/Akt in colon cancer cell line SW480.Methods When colon cancer cell line SW480 was culfured,Western blot was used to detect the expression of survivin protein in SW480 cells after they were stimulated by different concentrations of IGF-I for different lengths of time.The phosphorylation of Akt in SW480 cells was determined by Western blot after SW480 cells were stimulated by 100 ng/mL IGF-I with in 2 h.Before and after the specific inhibitor LY294002 was treated in SW480 cells to block the pathway of PI-3K/Akt,the phosphorylation level of Akt protein was detected by western blot,survivin protein by western blot and cell immunofluorescence and the cell apoptosis rates of different treated-groups were measured by flow cytometry.Results IGF-I up-regulated the expression of survivin in a dose-dependent and time-dependent manner in colon cancer line SW480(when treated with IGF-I for 12h,the expression of Survivin reached the peak and when treated with 100ng/ml IGF-I,the Survivin reached the highest expression level);IGF-I actived the signaling pathway of PI-3K/Akt of SW480 cells quickly(the expression of p-Akt proteins in SW480 reached the maximum level when cultured with IGF-I for 15 and 30min,but after that they quickly decreased);IGF-I induced survivin expression through the PI-3K/Akt in colon cancer line SW480;IGF-I could inhibited the apoptosis of SW480 cells through the PI-3K/Akt[use of flow cytometry for detection in the blank,stimulation and block groups showed the SW480 cell apoptosis rate was(5.18?0.415)%,(0.85?0.052)%,(3.15?0.411)% respectirely,with significant differences between the groups(P

ABSTRACT

Objective To investigate the ultrastructural features and it's significance of the skip area with giant cell arteritis(GCA).Methods 20 GCA patients and 7 non-GCA patients underwent temporal artery biopsies. The sections were stained with HE and methylene blue(MB) for light microscopy(LM), doubly stained with uranyl acetate and lead curate for electron microscopy (EM). 20 GCA patients were divided into skip area GCA group(14 cases) and active inflammatory cell infiltration GCA group(6 cases) according to the definition of skip area or skip lesion. The ultrastructural changes of each group were observed by EM and assessed by using 4 grades from 0 to 3 scores.Results The scores of ultrastructural changes were as follows: the skip area GCA group, 0 in 1 patient, 2 to 6 in 9 patients, and 8 to 9 in 4 patients; the active inflammatory cell infiltration GCA group, 7 to 15 in 6 patients; the non-GCA group, 0 to 1 in 6 patients, and 5 in 1 patient. The scores of the three groups were tested by H check, the difference was evidently significant( P0.05) in comparison with active inflammatory cell infiltration GCA group.Conclusions At electron microscopic level, the skip area of temporal artery biopsy with GCA showed obvious pathological changes, especially in the intima of artery. The results suggest that the investigation of ultrastructure features of the skip areas of temporal artery with GCA may be benefit for the diagnosis and treatment of this disease.
